The challenge tells you to kill all the targets, and that is the ultimate goal. However, the catch is that the challenge ends as soon as you defeat all four. If you zero in on just the targets (aka the people with the green markers), you’ll miss out on all the other enemies prowling the area. So, here’s the basic strategy – leave at least one target alive, and beat them only after you’ve cleared out all the other enemies.

Incidentally, the same strategy applies for completing the other challenge, fall damage kills. You won’t be able to do that if you zero in on the targets and nothing but, you won’t scrounge up enough overall points in the category to earn the gold medal.
